There's a number of Game Express, Gamestops, and EB's scattered everywhere from Soho-ish, Midtown, the 'outer' boroughs, etc. Multimedia is one-of-a-kind shop in St. Marks.
Here is what I do and recommend every time:
Go to www.yellow.com
Enter city/state whatever you want to narrow it down to.
Enter a category. Video Games.
Or Video Rental, or thrift, whatever.
Then it should give you eveywhere having to do with that subject with the city or entire state. You should have no problem then, making a list.

Carrying all imports and rare stuff you won't find in Gamestop and EB.
Around Chinatown, check many other games stores too that carry odd items, sometimes you find rare items in the stack.
It's funny... I was just in NYC the last 3 days and I found a GS and EB. I wanted to go to Multimedia 1.0 (As I've ordered from them online before) but it was too far away.

Don't get too excited. From my experience of living there a few years, NYC sucks for game shopping. I do better just searching in small cities here in Washington state.
The flea markets in NYC are visited by a TON of people, so it's almost pointless to go. Yard/Garage sales are out because most people don't even have yards or garages (nor do sensible people trust attracting shifty strange people to their homes). :/ The few independent stores/chains I've come across have terrible selections, even worse prices, and the people who visit/work them look way too shifty.
I've only done my game shopping at GameStop in NYC, sadly. That and online for everything else. As someone who loves to go out and hunt, that kills me. >_< Thankfully I got the summer to do hunting elsewhere.
